In Vitro Shelf-life for Antifungal Effect of Nano chitosan Integrated with Tissue Conditioner on Candida albicans.

Abstract

Background: The increasing number of elderly individuals worldwide, including Iran, resulted increasing of the edentulous population. Using dental prosthesis including complete denture is a common treatment method, which caused denture stomatitis. Tissue conditioner that used to improve the fit and comfort of dentures resulted more microbial especially candida colonization. The aim of present study is to evaluate the in vitro shelf-life of Chitosan Nanoparticles in combination with tissue conditioner on control of Candida albicans. Methods: Nano chitosan was developed and added to discs of tissue conditioner in the current experimental study and used to perform its antifungal susceptibility properties using disk agar diffusion and broth micro dilution susceptibility tests during 1 to 7 days and MFC and MIC90 were determined. Data was analyzed using SPSS20 software. A significance level of 0.05 was applied. Results: The prepared Nano chitosan was first analyzed and verified by Fourier-transform infrared spectroscopy and electron microscope. Only disks containing 5% and 2.5% Nano chitosan inhibited Candida albicans growth in 24 and 48 hours. There weren't any significant statistical differences between nystatin with 2.5 and 5% Nano chitosan during 1 to 2 days. The antifungal properties of Nano chitosan dramatically decreased over time and after 2 days there wasn't any antifungal effect. Conclusions: Our results in the present study showed that the addition of chitosan Nanoparticles to tissue conditioner can completely control the colonization of C. albicans in the first 2 days.
